IMMUNOgen - Immunoassays Plasma,Serum Anti-TPO Tests per Package: 100
IMMUNOgen The Eclectica Anti-TPO assay for the Eclectica and Eclectica TiCA analysers has been designed for the quantitative determination of antithyroid peroxidase (TPO) antibodies in human serum and heparinized plasma. The method can be used to measure Anti-TPO concentrations up to 5000 IU/ml of Anti-TPO. Primary calibration is against the WHO 1st International Reference Preparation (WHO 1st IRP 66/387) for Anti-TPO. Code: 1707100 Package: 2 x 50

The Eclectica Anti-TPO assay for the Eclectica analyzers has been designed for the quantitative determination of antithyroid peroxidase (TPO) antibodies in human serum and heparinized plasma. Primary calibration is against the WHO 1st International Reference Preparation (WHO 1st IRP 66/387) for Anti-TPO.

Thyroid Peroxidase (TPO) is a 110 kD membrane bound hemo-glycoprotein with a large extra cellular domain, and a short trasmembrane and intracellular domain. TPO is involved in thyroid synthesis and the apical pole of the follicular cell. Several isoforms related to differential splicing of TPO RNA have been described. TPO molecules may also differ respect to their three-dimensional structure, extent of glycosilation and heme binding. Most of the TPO molecules do not reach the apical membrane and are degraded intracellularly.TPO autoantibodies were initially described as anti-microsomial autoantibodies (AMA) since they were found to react with crude preparations of thyroid cell membranes. The microsomal antigen was later identified as TPO When extracted from human thyroid tissue, TPO may be used as a crude membrane preparation or may be purified by different methods. The assay specificity may also differ because of contamination by other thyroid antigens, notably Tg and or variations in the three dimensional structure of TPO. The use of recombinant human TPO (rhTPO) eliminates the risk of contamination but does not solve the problem of the differences in TPO structure that depend upon the technique used to isolate TPO.

TPOAb is the most sensitive test for detecting autoimmune thyroid disease. TPOAb is typically the first abnormality to appear in the course of developing hypothyroidism secondary to Hashimoto’thyroiditis. In fact when TPOAb is measured by a sensitive immunoassay, >95 % of subjects with Hashimoto thyroiditis have detectable levels of TPOAb. Such methods also detect TPOAb in most (about 85%) patients with Graves’ disease. Patients detected in early pregnancy are at risk for developing post-partum thyroiditis.Patients with Down’s syndrome have an increased risk of thyroid dysfunction due to autoimmune thyroid disease and annual screening with TSH and TPOAb is important.

The Eclectica assays for the Eclectica analyzers consists of:

  1. Eclectica Anti-TPO Reagent Kit: specific reagents of Eclectica Anti-TPO kit to execute the test and to verify the standardization.
  2. Eclectica Anti-TPO Calibration Kit: for standardization.
  3. Eclectica Diluent 2 Kit: for samples dilution
  4. Eclectica Immunoassay Common Reagents Kit: for general chemistry reaction.
  5. Eclectica Immunoassay Wash Solution: Wash Solution specific for Immunological reaction.
  6. Eclectica Systemic Wash Solution: Wash Solution of Eclectica system.

In the Eclectica Anti-TPO assay two TPO-conjugates are used in an immunoenzymetric assay system which incorporates magnetic solid phase separation. All incubation times and reagent volumes are determined by an assay specific software protocol.

Fixed amounts of fluorescein-conjugated TPO and a TPO-conjugated to alkaline phosphatase are added to a sample, control, or standard. The reactants are incubated at 37°C. During the incubation fluorescein-TPO conjugate, Anti-TPO and an alkaline phosphatase TPO conjugate   forming a sandwich.

At the end of the incubation period anti-fluorescein coupled to a magnetic solid phase is added in excess. This rapidly and specifically binds to the Anti-TPO complex and is sedimented in a magnetic field.

After aspirating the liquid phase and washing the solid phase, a solution of the enzyme substrate, phenolphthalein monophosphate is added to the incubation cell and the each cell incubated at 37°C. After incubation the enzyme reaction is stopped by the addition of a stop reagent and the intensity of the color developed is measured photometrically. The intensity of the color developed is directly proportional, within the working range of the assay, to the concentration of Anti-TPO in the sample. The concentration of Anti-TPO in a patient sample or control is then determined by interpolation from a stored standard curve.
